When talking about my research experience, I am overwhelmed. I have participated in many projects, and each project brings me different feelings. The first time I did scientific research was the school's "Challenge Cup" research project. Although I got a lot of guidance from the teachers and the experimental process was relatively smooth, I did not encounter no problems. The biggest difficulty was the time-consuming nature of the experiment and the high concentration required during the antibody staining process. It takes a whole day to do one experiment, and each experiment has to do five different protocols at the same time, sometimes dozens of tubes of samples at a time, so you have to be very careful when doing the antibody staining, and the slightest negligence may lead to the end of your work. When I first started to do the experiment, because I was not familiar with the experimental operation, I often added the wrong reagent or the staining result was unsatisfactory, which led to the day's hard work being wasted. However, through repeated practice, I gradually mastered the operation skills and overcame these difficulties. Another project was my graduation design, which was also a challenging process. The experiment itself was not difficult; the biggest problem was the use of statistical methods. Through this project, I gained insight into the differences between different statistical methods and the problem of false positives. When analyzing the data, I found that the results were statistically different when analyzing the experimental and healthy control groups of CHI3L1 using the U test alone, but not in the Kruskal-Wallis multiple comparison test.
